Advanced Hypoimmune Human iPSCs for Allogeneic Cell Therapy Research

A ready-to-use iPSC platform designed to reduce immune recognition

Immune rejection remains a major challenge for allogeneic cell therapies. TARGATT™ RUO Hypo Plus Human iPSC (#AST-9750) from Applied StemCell (ASC) is a research-use-only human iPSC (hiPSC) line engineered to serve as a starting point for immune-evasion studies.
The B2M/CIITA double knockout eliminates HLA class I and class II expression, thereby reducing recognition by CD8+ and CD4+ T cells. In addition, TARGATT™, ASC’s proprietary site-specific integration technology, enabled the targeted integration of five immune-modulatory genes – HLA-F-B2M, HLA-G-B2M, CD200, CCL21, and SERPINB9 – at the H11 safe-harbour locus. These genes target complementary pathways involved in innate and adaptive immune regulation.

Key features of TARGATT™ RUO Hypo Plus Human iPSC:

  • B2M/CIITA double knockout: Eliminates HLA Class I and Class II expression, reducing recognition by CD8+ and CD4+ T cells.
  • Five immune-modulatory genes: HLA-F-B2M, HLA-G-B2M, CD200, CCL21, and SERPINB9 target complementary immune-regulatory pathways.
  • Quality-controlled hiPSC line: Pluripotency-marker expression (OCT4, SOX2, TRA-1-60, and SSEA4), normal karyotype, and pathogen- and mycoplasma-free status.

From cell-line engineering to functional research

Building a complex hypoimmune hiPSC line from scratch involves multiple engineering and validation steps. TARGATT™ RUO Hypo Plus Human iPSC (#AST-9750) provides a pre-engineered iPSC platform that allows researchers to focus on downstream applications, including differentiation strategies and functional studies.

The platform can be used to investigate allogeneic and multi-lineage immune-evasion strategies and to assess their potential in next-generation allogeneic cell therapies.
