Description

Tissue factor (TF) is a transmembrane protein that functions as the high affinity receptor for factor VIIa (FVIIa)¹. It is “sequestered” on the surface of cells not exposed to blood. Following vascular injury, formation of a TF/FVIIa complex (aka extrinsic tenase complex) initiates the hemostatic response by catalyzing the conversion of factor X to factor Xa and factor IX to factor IXa. Beyond its role in coagulation, evidence of a role for TF/FVIIa in cell signalling responses has also been observed^2,3.

TF consists of three domains⁴:  (1)  an extracellular domain, that serves as a template for factor VIIa binding; (2) a transmembrane domain; and (3) a cytosolic domain of 21 amino acids length inside the cell which is involved in the signaling function of TF. Soluble forms of TF lacking the transmembrane and cytoplasmic domains have been observed though their roles in promoting thrombin formation are not clear⁵.

Human (Full-Length) Recombinant Tissue Factor is produced in Sf9 cells and purified from cell extracts using conventional chromatography techniques. It is relipidated with PCPS vesicles. Purified HTF is supplied in 15 mM Tris, 15 mM NaCl, pH 7.4 containing 82 mM glycine, 5.9 mM NaN₃, 0.1% ovalbumin, and 115 mM trehalose•2H₂0 and should be stored at -80°C. Purity is determined by SDS-PAGE analysis and activity measured using a fluorogenic substrate.
