Functional Human Cryopreserved Platelets for Research Applications

Reliable access to functional human platelets remains a persistent challenge in platelet-focused research. Dependence on freshly drawn donor platelets introduces logistical complexity, high operational costs, and significant donor-to-donor variability—limiting reproducibility and scalability.

Retham™ Cryoplatelets are functional human cryopreserved platelets designed for Research Use Only (RUO) applications. Upon thawing, they retain responsiveness across multiple platelet activation pathways and assay formats, enabling consistent, flexible, and scalable research workflows.

 

Why Retham™ Cryoplatelets?
Traditional platelet sourcing presents key challenges:

– Logistical burden of donor recruitment and blood collection

Retham™ Cryoplatelets address these limitations by providing a ready-to-use, standardized platelet source.

Product Features

– Human platelets derived from healthy donors

– Blood group O pooled platelets

– Three-donor pools per lot for reduced variability

– Cryopreserved for on-demand use

– Compatible with a wide range of functional platelet assays

Functional Performance
General workflow:

– Thaw at 37°C (~3 minutes)

– Remove cryoprotectant with centrifugation

– Resuspend in an appropriate activation buffer/normal plasma

– Stimulate with agonists

– Measure functional readouts

 

Granule Release Assays

Retham™ Cryoplatelets demonstrate functional responses in granule release formats, including:

– Responsiveness to TRAP (Thrombin receptor activating peptide)

– Intact FcGRIIa (CD32)-mediated signaling as evidenced by antibody-mediated activation

 

Aggregation-Based Assays
Demonstrated responsiveness to multiple agonists:

– TRAP

– Collagen

– Ristocetin

– ADP

– Epinephrine

– Arachidonic Acid

While higher agonist concentrations may be required versus fresh platelets, Retham™ Cryoplatelets offer broad agonist responsiveness, unlike many lyophilized alternatives.

Procoagulant Activity
Retham™ Cryoplatelets support robust coagulation-based assays:

– Thrombin Generation (CAT): ~2x higher thrombin generation vs. fresh platelets

– Thromboelastography: Strong clot formation

– Shortened lag phase observed across assays

These characteristics suggest a procoagulant phenotype, potentially reflecting partial activation or altered membrane dynamics post-cryopreservation.
