MAD7 Gene Editing

While CRISPR/Cas9 transformed modern biology, its complex licensing model often requires agreements with multiple organizations.

Fortunately, MAD7 was introduced by Inscripta, Inc. in 2017 to provide the scientific community with broader access to advanced genome editing technology.

How MAD7 Compares to Cas9

MAD7 (ErCas12a) is an RNA-guided nuclease with distinct functional benefits over standard Cas9:

  •  Single-RNA System: Requires only one RNA species to target DNA sequences.
  • Sticky-End Cleavage: Produces staggered double-strand breaks instead of blunt ends.
  • Expanded PAM Diversity: Targets T-rich PAM sequences (5′-TTTN-3′ / 5′-CTTN-3′) rather than G-rich sites (5′-NGG-3′).

MAD7-based gene editing is backed by years of validated success in hiPSC lines and HEK293 cells.
